Dual esterase activity may be identified by combined methods. This preparation of bone marrow from a patient with AMML first was incubated in a mixture containing buffer, fast blue RR salt, and alpha-naphthyl acetate then was incubated in another mixture of fast garnet and naphthol AS-D chloracetate (naphthol AS-D chloracetate esterase is found specifically in granulocytes). The monocytes in this bone marrow contain blue granules; the granulocyte granules stain red. Some of the cells, including some of the segmented neutrophils, contain both blue and red granules.
