This is the bone marrow preparation of a patient with AMML tested for nonspecific esterase activity in monocytes with anaphthyl butyrate as substrate and fast red PDC salt for coupling. Both alpha-naphthyl acetate and alpha-naphthyl butyrate are excellent for identifying the esterase activity in monocytes. The monocytes have numerous red-staining granules, while all other cell types are negative. This reaction also is inhibited by fluoride. The substrates, stains, buffers, and procedures are available in kits from Sigma Chemical Co, St. Louis.
